BTF3a/b Antibody (H-9) is a mouse monoclonal IgG2b antibody that detects BTF3 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-BTF3 antibody (H-9) recognizes two isoforms of BTF3, BTF3a and BTF3b, which are produced from alternative splicing of the BTF3 gene. While both isoforms share a high degree of similarity, BTF3b is characterized by the absence of the first 44 amino acids at the N-terminal of BTF3a, which renders BTF3b incapable of inducing transcription despite binding to RNA polymerase II. This distinction is crucial as BTF3a plays a significant role in the initiation of gene transcription by forming a stable complex with RNA polymerase II, thereby facilitating the assembly of the transcription initiation complex at the TATA box. Furthermore, both isoforms interact with protein kinase CK2, which is essential for the phosphorylation of BTF3a and TFIIB, ultimately influencing the transcription efficiency of tRNA and 55 rRNA genes by RNA polymerase III.
