BTF3a/b Antibody (C-7) is a mouse monoclonal IgG1 antibody that detects BTF3 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). BTF3 plays a crucial role in the initiation of gene transcription by forming a stable complex with RNA polymerase II, thereby facilitating assembly into the transcription initiation complex. This process is vital for accurate expression of genes, particularly those involved in tRNA and rRNA synthesis, which are essential for protein translation and cellular function. The BTF3 gene encodes two isoforms, BTF3a and BTF3b, produced through alternative splicing, with BTF3a being the active form that can induce transcription. BTF3a is phosphorylated by protein kinase CK2, which enhances transcriptional activity, while BTF3b, lacking the first 44 amino acids of BTF3a, cannot induce transcription despite binding RNA polymerase II. These isoforms and their interaction with CK2 underscore the complexity of transcription regulation and highlight BTF3′s importance in maintaining proper gene expression.
