basonuclin Antibody (1F4) is a mouse monoclonal IgG2b kappa light chain antibody that detects basonuclin protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-basonuclin antibody (1F4) is available as a non-conjugated format. basonuclin protein, a zinc finger transcription factor, plays a crucial role in regulating ribosomal DNA transcription, which is essential for ribosome biogenesis and protein synthesis in cells. basonuclin expression is highly restricted to specific tissues, being predominantly found in keratinocytes of the basal layer of the epidermis, the outer sheath of hair follicles, and in germ cells of the testis and ovary. basonuclin is associated with chromatin throughout the cell cycle, including during mitosis, but is absent in post-mitotic cells, indicating importance in cell proliferation and differentiation. In the epidermis, basonuclin is primarily localized in the cytoplasm but translocates to the nuclei of basal cells during various stages of keratinocyte growth, suggesting a role in regulating gene expression during cellular development. By enhancing rRNA synthesis through interaction with rDNA promoters and modulating RNA polymerase I activity via zinc finger domain, basonuclin may serve as a cell-type-specific transcription factor, making basonuclin a significant player in cellular growth and function.

I am using basonuclin (1F4): sc-517114 for immunofluorescence, what dilution of this antibody should I use?

Asked by: Trav11
Thank you for your question. We recommend a starting dilution of 1:50. However, optimal antibody concentration should be determined by titration. Please find our full immunofluorescence protocol here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
