BANP, also known as BTG3 associated nuclear protein, BEND1, SMARBP1, or SMAR1, is a crucial 519 amino acid protein that resides in the nucleus and is a member of the BANP/SMAR1 family. BANP plays a significant role in regulating V(D)J recombination during T-cell development by interacting with key proteins such as CDP and p53. Specifically, BANP binds to a scaffold/matrix DNA attachment region, which is essential for repressing the enhancer function of the T-cell receptor beta (TCRβ). Furthermore, BANP is involved in promoting the phosphorylation of p53 at serine residues, leading to the nuclear accumulation of p53 and subsequent cell cycle arrest, highlighting BANP′s importance in cellular regulation and tumor suppression. Notably, BANP expression is downregulated in breast cancer cells, suggesting a potential role as a tumor suppressor. Additionally, three isoforms of BANP arise from alternative splicing events, which may contribute to diverse functional roles in cellular processes.
