Band 3 Antibody (BIII 136) is a mouse monoclonal IgG2a antibody that detects band 3 in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Band 3, also known as AE1, is a crucial erythrocyte membrane glycoprotein that plays a vital role in maintaining cell structural integrity and facilitating the exchange of chloride and bicarbonate ions across the phospholipid bilayer. Band 3 consists of approximately 900 amino acids and features two distinct domains that mediate diverse functions. The amino terminal domain, referred to as the cytoplasmic domain of erythrocyte membrane band 3 (cdb3), serves as an attachment site for the erythrocyte cytoskeleton by binding to ankyrin, which is essential for maintaining the shape and stability of red blood cells. The carboxy-terminal, membrane-associated domain is responsible for the anion exchange transport, a critical process for maintaining acid-base balance in the blood. Notably, band 3 degradation can lead to the formation of an aging antigen known as senescent cell antigen (SCA), which is expressed on older cells and signals their removal by the immune system, highlighting band 3′s importance in cellular aging and immune response. Additionally, a band 3 isoform that lacks the first 65 amino acids and does not bind ankyrin is expressed in the kidney, indicating functional diversity across different tissues.
