Bag-5 Antibody (18Z) is a mouse monoclonal IgG1 antibody that detects Bag-5 in human samples through applications such as western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Bag-5, also known as Bcl-2-associated athanogene 5 or Bag family molecular chaperone regulator 5, is a crucial member of the Bag family of proteins, characterized by four Bag domains. Anti-Bag-5 antibody (18Z) plays a significant role in studying cellular stress responses by binding with high affinity to the HSC 70/HSP 70 ATPase domain, thereby regulating chaperone activity and apoptosis. Notably, Bag-5 is a key component of the HSP 70/Parkin complex, where Bag-5 inhibits the E3 ubiquitin ligase activity of Parkin and modulates HSP 70 chaperone activity. The interaction between Bag-5 and the ATPase domain of HSP 70, as well as the N-terminal linker region of Parkin, is vital for maintaining cellular homeostasis, particularly in the context of dopaminergic neuron injury. Upon such injury, Bag-5 expression is upregulated, sensitizing neurons to cell death, which underscores Bag-5′s potential as a therapeutic target in neurodegenerative diseases like Parkinson′s disease, where mutations in the Parkin gene are implicated. Bag-5 monoclonal antibody (18Z) is an invaluable tool for researchers investigating the molecular mechanisms underlying these critical cellular processes.
