The Atto 565 NHS ester is a frequently employed reagent that reacts with primary amines. This amine-reactive compound is extensively utilized for protein labeling, enabling the formation of a stable amide bond between the dye and amino groups in the target molecule, such as proteins. The optimal pH range for efficient coupling using NHS ester is between 8.0 and 9.0. Within this pH range, the amino groups, specifically the ε-amino groups of lysines in proteins, are sufficiently deprotonated to facilitate rapid coupling reactions.
