ATM Kinase Inhibitor (KU 55933) is a cell-permeable, potent, selective and ATP-competitive inhibitor of ATM (Ataxia telangiectasia mutated), a serine/threonine protein kinase, that exhibits an IC50 of 13 nmol/L and a Ki of 2.2 nmol/L. ATM is rapidly and specifically activated in response to DNA double-strand breaks in eukaryotic cells and signals to cell cycle and DNA repair components by phosphorylating downstream targets such as p53, CHK2, NALP2 (NBS1), and BRCA1. Investigators discovered that this product shows inhibition specificity for other phosphatidylinositol 3#39-kinase-like kinases as well. Research shows that HIV-1 integrase stimulates ATM-dependent DNA damage response and that this inhibitor is capable of suppressing the replication of both wild-type and drug-resistant HIV-1. Studies also reveal two novel ATM functions: regulation of ribonucleotide reductase and mitochondrial homeostasis. Additional research shows that ATM Kinase Inhibitor and ATM/ATR Kinase Inhibitor (sc-202964) induces senescent breast, lung, and colon carcinoma cells to undergo cell death which is directly effected by p21.
