Amylin Antibody (E-5) is a mouse monoclonal IgG2a kappa light chain antibody that detects Amylin protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-Amylin antibody (E-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatility for different experimental needs. Amylin, also known as Islet Amyloid Polypeptide (IAPP), is a 37-amino acid polypeptide that plays a crucial role in glucose metabolism and is secreted by pancreatic beta cells alongside insulin. Amylin′s primary function is to regulate glucose levels by slowing gastric emptying and promoting satiety, which is particularly important in the context of diabetes, as dysregulation of Amylin can contribute to the pathophysiology of type 2 diabetes. Amylin forms amyloid fibrils in the pancreas of diabetic patients, which can lead to beta-cell dysfunction and loss, highlighting Amylin′s significance in both normal physiology and disease states. The gene encoding Amylin is located on human chromosome 12p12.3-p12.1, and Amylin expression is tightly regulated, making Amylin a valuable target for research into metabolic disorders.

In pdf i saw (sc-377530), amylin band between 10k-16k, and molecular weight is 4kDa. Kindly clarify on this.

Thank you for your question. The western blot shows lysates from cells that were transfected with recombinant protein (target protein+marker). The protein itself has a molecular weight of approximately 4 kDa.

What do you mean by the antibody detecting "Amylin precursor and active peptide"? Does this refer to monomeric or oligomeric amylin?

Thank you for your question. This monoclonal antibody is raised against amino acids 40-89 of the full length human protein, which, according to Uniprot is processed into a Islet amyloid polypeptide, corresponding to amino acids 34-70. Therefore, we expect the antibody to recognize both precursor and active processed forms of Amylin. https://www.uniprot.org/uniprot/P10997#ptm_processing

How specific is the antibody for human amylin? Is there any cross reactivity for amylin of other species (especially rat)?

Thank you for your question. We have not yet completed testing of this antibody, Amylin (E-5): sc-377530, for reactivity in other species including mouse and rat. The antibody is raised against an antigen sequence derived from human Amylin, however that sequence does share ~ 70% sequence identity with the corresponding protein in mouse and rat. The exact epitope is not known for this antibody, but there is potential for cross-reactivity with these other species.

Hi, l would like to use your Amylin antibody E-5 and H-50 as bioreceptors in my Biacore assay, to detect Amylin as analyte based on antigen-antibody binding. Could you please send me the links of Amylin that I should I buy? Thank you very much in advance!

Thank you for your question. We have not tested any of our antibodies using the Biacore assay and so we are unable to offer you any specific support using the antibodies in this system. We do not currently sell any Amylin protein or peptide that could be used as a synthetic antigen with our Amylin antibodies.

Where I'll see the band of amylin antibody (sc-377530) in western blot, after transfecting HEK 293T cells with IAPP plasmid (bp- 5689)? Will I see band near 4kd or between 10-16k as shown in your pdf.
