Aminoglutethimide has been shown to inhibit human placental CYP19 (aromatase) in vitro. In ovine adrenocortical cells, aminoglutethimide has been shown to inhibit ACTH receptor mRNA expression in a time-dependent manner in vitro. It significantly suppresses baseline ACTH-R mRNA expression and steriod secretion in a dose-dependent manner in vitro in the human NCI-h295 adrenocortical carcinoma cell line. In vivo, Aminoglutethimide is known to accelerate its own metabolism and the metabolism of synthetic dexamethasone and glucocorticoid.

Khan et alKhan et al. (PubMed ID 26102013) foudn that aminoglutethimide peroxidative metabolism led to the down-regulation of critical anti-apoptotic proteins responsible for inhibiting the release of pro-apoptotic factors from the mitochondria as well as cytoskeletal proteins such as nuclear lamina. -SCBT Publication Review
