ALDH1L2 encodes a mitochondrial aldehyde dehydrogenase that functions as a 10-formyltetrahydrofolate dehydrogenase, supporting one-carbon folate metabolism and redox homeostasis within mitochondria. By regulating formyl-THF turnover and NADP(H)-linked reactions, ALDH1L2 influences nucleotide biosynthesis capacity, mitochondrial oxidative balance, and cellular responses to metabolic stress. Perturbation of mitochondrial one-carbon flux can reshape anabolic demands and reactive oxygen species handling, processes frequently altered in proliferative and stress-adapted cell states. Accordingly, ALDH1L2 is studied in the context of metabolic rewiring, mitochondrial function, and disease-associated changes in folate-dependent pathways.
ALDH1L2 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the ALDH1L2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the ALDH1L2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the ALDH1L2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish ALDH1L2 protein expression.
This CRISPR knockout system enables efficient generation of ALDH1L2-deficient cell models for investigation of ALDH1L2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
