SEE ALSO...

AHNAK Antibody (E-5) is a mouse monoclonal IgG2a kappa light chain antibody that detects AHNAK protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). AHNAK (E-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. AHNAK protein, also known as neuroblast differentiation-associated protein AHNAK or desmoyokin, is a large 5,890 amino acid protein encoded by the intronless human gene AHNAK located on chromosome 11q12. AHNAK features a complex structure with three main regions: a 251 amino acid N-terminus, a substantial central region comprising 4,390 amino acids with multiple repeated units, and a 1,002 amino acid C-terminus. The central region is characterized by antiparallel beta-strands linked by loops, which is crucial for structural integrity and function. The C-terminal region contains several putative regulatory elements, including nuclear export localization signals, a leucine zipper, and potential phosphorylation sites for key kinases such as Akt1 and protein kinase C. These features suggest AHNAK plays a significant role as a signaling molecule in various physiological processes, including neuronal cell differentiation and modulation of cardiac L-type calcium channel function in response to β-adrenergic stimulation.

Hello! I plan to use AHNAK Antibody (E-5) for Western blot from the whole kidney lysates. Could you please suggest optimal running conditions (non-reducing vs reducing) and transfer conditions? Thank you!

Asked by: Dr Valeria
Thank you for your question. Please follow electrophoresis directions indicated by the manufacturer or your electroblotting apparatus, we can not provide these conditions. You can find our recommended Western blot protocol here: https://www.scbt.com/resources/protocols/western-immuno-blotting

I am using AHNAK (E-5): sc-390743 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Dr Ninau Qelp
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 4 out of 5 by from Seemed good for western blottingI used mouse Hepa1-6 cell lysates with loading gradient to check this antibody in western blotting. Antibody was diluted 1000X and incubated at 4℃ overnight. The exposure results showed one single band at around 160KD. But I didn’t use si specific genes to check whether this band was specific.
