ADP-ribose-pNP functions as a colorimetric substrate to evaluate the activity of poly(ADP-ribose)polymerase (PARP) enzymes. The absorption of the released p-nitrophenol is measured at 405 nm, and the slope of the calibration curve is utilized to convert the absorbance values into moles of generated product. Utilizing the ADP-ribose-pNP-based continuous assay offers significant advantages over conventional discontinuous PARP assays, facilitating high-throughput screening of PARP-1, tankyrase-1, and VPARP activities, as well as the evaluation of their inhibitors.
