Ac-YVAD-AFC is a fluorogenic substrate for caspase-1 and caspase-4. Ac-YVAD-AFC functions by specifically targeting caspase-1, a enzyme involved in the inflammatory response. Caspase-1 plays a significant role in cleaving pro-inflammatory cytokines, including interleukin-1β and interleukin-18. By binding to the active site of caspase-1, Ac-YVAD-AFC hinders the cleavage of pro-inflammatory cytokines, thereby mitigating inflammation. Its effects extend to diverse biochemical and physiological aspects, encompassing the inhibition of pro-inflammatory cytokine production like interleukin-1β and interleukin-18, as well as the suppression of other inflammatory mediators such as nitric oxide and prostaglandin E2.
