Ac-IETD-AFC is a potent inhibitor of protein synthesis. It operates by binding to the peptidyl transferase center of the ribosome, effectively impeding the peptidyl transferase reaction necessary for protein synthesis. Additionally, Ac-IETD-AFC binds to the A-site of the ribosome, responsible for the attachment of aminoacyl-tRNA molecules. This binding obstructs the interaction of aminoacyl-tRNA molecules, preventing peptide bond formation and subsequent elongation of the polypeptide chain. Consequently, the inhibition of protein synthesis by Ac-IETD-AFC hampers cell growth, differentiation, and apoptosis. Ac-IETD-AFC has demonstrated the ability to inhibit the translation of specific mRNA molecules, leading to the suppression of distinct cellular processes.
