    SEE ALSO...

AATK Antibody (L-37) is a mouse monoclonal IgG1 kappa light chain antibody that detects AATK protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-AATK antibody (L-37) is available as the non-conjugated form. AATK, or Apoptosis-associated tyrosine kinase, also known as LMR1 (Lemur tyrosine kinase 1), AATYK, AATYK1, or LMTK1, is a single-pass type I membrane protein primarily localized in the brain, where AATK plays a critical role in neuronal differentiation and apoptosis. AATK′s importance lies in its function within death activation pathways, which are essential for regulating neuronal apoptosis—a process that minimizes brain damage and ensures proper neural development. AATK expression is induced during apoptosis, suggesting involvement in growth arrest of myeloid precursor cells. Structurally, AATK features a proline-rich domain at its C-terminus and a tyrosine kinase domain at its N-terminus, which contribute to in vitro kinase activity. Notably, three isoforms of AATK exist due to alternative splicing events, allowing for functional diversity in cellular processes.

why the size of data sheet for this antibody is 145 kDa but the western blot picture showed from 86-110 kDa ? any explanation ?

    Asked by: ody82
Thank you for your question. According to Uniprot (see below), AATK has 3 isoforms with predicted molecular weights of 145 kDa, 45 kDa and 93 kDa. This antibody would not be able to recognize isoform 2 (missing amino acids 1-433). Based on the observed molecular weights alone, this antibody may recognize both isoform 1 and 3 in the cell lines tested. https://www.uniprot.org/uniprot/Q6ZMQ8#sequences

Can sc-100436: AATK (L-37) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

    Asked by: Professor Griffin
Thank you for your question. Yes, sc-100436: AATK (L-37) is recommended for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 4 out of 5 by from Good in WB, questionable species cross reactivityWorks well in untransfected HEK293 lysate, single clean band of the correct size when used at 1:250 in 1% milk. However, nothing in whole brain lysate. LMTK1 is clearly enriched in neurons as per multiple literature citations. Maybe the antibody does not cross react with mouse and only recognizes human protein?
