5-Bromo-4-chloro-3-indolyl β-D-galactopyranoside (X-Gal) is a histochemical substrate for beta-galactosidase. 5-Bromo-4-chloro-3-indolyl β-D-galactopyranoside is cleaved by beta-galactosidase to yield an insoluble blue preceipitate. It is used to detect DNA insertions into the lacZ region of plasmid DNA, which causes the loss of beta-galactosidase activity. It is also used in conjunction with IPTG for detection of b-Galactosidase activity in bacterial colonies in a colorimetric assay.
