6-(N-Trifluoroacetyl)caproic acid NHS, also known as TCFS, is an amine-reactive modification reagent with a protected primary amine group. It forms amide bonds with primary amines (e.g., lysine side chains) to add a seven-atom spacer arm (7.7 angstroms) terminated by a trifluoroacetyl group that can be removed later to expose a new primary amine. Its benefits lie in its ability to add a protected primary amine group, extend lysine side-chain length to alter steric hindrance and block amine groups and replace them with one on a 7-atom spacer. The leaving group is readily removed unmasking a primary amine capable of reacting with any amine-reactive crosslinker.
