6-(Methylsulfinyl)hexyl isothiocyanate, found in wasabi (W. japonica), exhibits a range of diverse biological activities. Initially identified as a glycogen synthase kinase-3β inhibitor, it has demonstrated inhibitory effects on various processes. At a concentration of 5 µM, it effectively reduces nitric oxide (NO) production in mouse peritoneal exudate macrophages. Furthermore, it hinders platelet aggregation induced by arachidonic acid in isolated human platelets (IC50 = 21.9 µM). In cancer cell lines encompassing breast, brain, colon, lung, ovarian, renal, and prostate cancer, 6-(Methylsulfinyl)hexyl isothiocyanate decreases cell survival (mean IC50 = 43.7 µM). In a mouse model of Parkinson′s disease induced by 6-OHDA, the administration of 6-(Methylsulfinyl)hexyl isothiocyanate (5 mg/kg twice weekly) diminishes apoptosis of substantia nigral dopaminergic neurons and mitigates motor dysfunction. Additionally, it reduces the occurrence of lung metastases in a B16/F10 murine melanoma model. Notably, 6-(Methylsulfinyl)hexyl isothiocyanate inhibits viability in PANC-1 and BxPC-3 cells and acts as an inhibitor of the expression of the CSC signaling molecule SOX2.
