5-Bromo-4-chloro-3-indolyl α-D-galactopyranoside, referred to as X-Gal, is a chemical compound used in molecular biology techniques to detect the activity of the enzyme β-galactosidase. It is a colorless or pale yellow powder that, when cleaved by β-galactosidase, produces a blue precipitate or color change. 5-Bromo-4-chloro-3-indolyl α-D-galactopyranoside is used in assays such as blue-white screening and lacZ reporter gene assays. In these assays, 5-Bromo-4-chloro-3-indolyl α-D-galactopyranoside is added to a medium or substrate to detect the presence and activity of β-galactosidase, which indicates the expression of a target gene or the presence of a recombinant plasmid. Upon cleavage by β-galactosidase, 5-Bromo-4-chloro-3-indolyl α-D-galactopyranoside is hydrolyzed to yield 5-bromo-4-chloro-3-hydroxyindole, which spontaneously dimerizes to form a blue compound. This blue coloration or precipitate can be visually detected or quantified using spectrophotometric methods.
