4-Methylumbelliferyl α-L-idopyranosiduronic acid is a fluorogenic substrate for α-L-iduronidase. Active α-L-iduronidase cleaves the Methylumbelliferyl moiety from the synthetic substrate, which results in release of a fluorescent signal. The 4-methylumbelliferone core is a widely used fluorophore that emits blue light when excited by ultraviolet (UV) light, but its fluorescence is quenched when attached to the α-L-Idopyranosiduronic acid. The reaction is stopped by the addition of sodium carbonate.
