Josamycin is a member of the leucomycin group of macrolide antibiotics with a 16 member lactone ring produced by Streptomyces kitasatoensis and Streptomyces narbonensis var. josamyceticus. The compounds main use has been against gram-positive bacteria. The compound has been documented to function through direct inhibition of peptidyltransferase reaction as well as drop off of peptidyl-tRNA translation from ribosomes. The strength action was observed to be dependent on peptide length, no more than 3 amino acids, and composition of amino acids, fMet-Phe-tRNA or fMet-Val-tRNA. Kinetic studies demonstrated that Josamycin binds directly to the ribosome.
