Novobiocin Sodium Salt is a small molecule inhibitor of eukaryotic Topo II (DNA topoisomerase II) and prokaryotic DNA gyrase. In the case of DNA gyrase inhibition, Novobiocin functions as a competitive inhibitor of the ATP binding event necessary for turnover of the enzyme after a manipulation of DNA supercoils. Inhibition of these enzymes blocks the mechanism for detangling DNA and establishing the proper super-coiled state necessary for normal replication and transcription, compromising the viability of the cell. Cell treatment with Novobiocin Sodium Salt is shown to potentiate the cytotoxic effects of DNA alkylating agents. Novobiocin Sodium Salt is also described to inhibit DNA Polymerase (retroviral RNA-dependent DNA polymerase). Novobiocin Sodium Salt is an inhibitor of Hsp90 and ABCG2. Novobiocin Sodium Salt interacts with the C-terminal ATP-binding domain of Hsp90.
