NS3694 also known as Apoptosis Inhibitor II is a cell-permeable diarylurea compound that suppresses the formation of the apoptosome Apaf-1. Studies suggest that the main component to signal the activation of apoptosis is the release of mitochondrial proapoptotic proteins into the cytosol which leads to the activation of caspases which form the apoptosome. NS3694 inhibits the formation of the apoptosome Apaf-1 by blocking the activation of the initiator caspase-9. However, NS3694 does not exhibit any effect on apoptosome-independent caspase activation and enzymatic activity of caspases.
