Thiacetazone is a topical antimicrobial agent that inhibits the growth of microorganisms through the modification of mitochondrial membrane potential. This compound exerts its inhibitory effect on mycolic acid cyclopropane synthase. Thiacetazone prevents the conversion of double bonds within mycolic acid precursors into cyclopropyl rings in Mycobacterium bovis var BCG, M.marimum, and M.chelonae. This disruption is done by affecting the cyclopropyl mycolic acid synthases (CMASs), inducing the accumulation of unsaturated mycolate precursors.
