22(R)-hydroxy Cholesterol is an endogenous agonist for the oxysterol receptor, LXR (liver X receptor) which upon activation stimulates epidermal differentiation and development while inhibiting keratinocyte proliferation. LXRs also regulate the metabolism of cholesterol by increasing the transcription of the ABCA1 gene, which promotes cholesterol efflux and HDL formation. In addition, 22(R)-hydroxy cholesterol produces an elevation in CYP3A mRNA concentrations which is mediated by the pregnane X receptor (PXR). Studies suggest that 22(R)-hydroxy cholesterol promotes the transcription of the CYP7A1 gene via activation of LXRalpha receptors in rats. Furthermore, CYP7A1 activity promoted by LXRalpha can be inhibited downstream by FXR.
