    Overview

    PSMB5 encodes the β5 catalytic subunit of the 20S core proteasome, which provides the chymotrypsin-like activity essential for ATP-dependent protein degradation by the ubiquitin–proteasome system. This proteolytic pathway regulates protein quality control, antigen processing for MHC class I presentation, and turnover of key cell-cycle and stress-response regulators. Altered proteasome function has been linked to dysregulated proteostasis, oxidative and ER stress signaling, and changes in NF-κB and apoptotic pathways. PSMB5 is also studied in contexts such as proliferative signaling and responses to proteasome inhibition, where β5 activity can influence cellular sensitivity and resistance phenotypes.

    20S Proteasome β5 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the PSMB5 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the PSMB5 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.

    The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the PSMB5 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish 20S Proteasome β5 protein expression.

    This CRISPR knockout system enables efficient generation of PSMB5-deficient cell models for investigation of 20S Proteasome β5 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
