2-Hydroxypropyl-β-cyclodextrin is the most widely used modified cyclodextrin. 2-Hydroxypropyl-β-cyclodextrin has shown to change the physicochemical properties of lipophilic compounds when co-administered. 2-Hydroxypropyl-β-cyclodextrin functions by forming an inclusion complex with 2-Hydroxypropyl-β-cyclodextrin being administered for easier diffusion across biological membranes. The popularity of 2-Hydroxypropyl-β-cyclodextrin can be attributed to its large 7 glucose unit cavity size. Some advantageous effects are reduction of negative effects, increased aqueous solubility and increased stability. The effects of 2-Hydroxypropyl-β-cyclodextrin have been observed to be dose dependent with both advantageous and disadvantageous results occurring at sporadic concentrations. 2-Hydroxypropyl-β-cyclodextrin may also increase the antimicrobial effectiveness of chemical agents by increasing their release rate. 2-Hydroxypropyl-β-cyclodextrin is also known as Hydroxypropyl-beta-cyclodextrin, (2-Hydroxypropyl)-beta-cyclodextrin, 2-hydroxypropyl ethers β-cyclodextrin, and HP-β-CD.

The methodology for solubilizing a lipophilic compound in an aqueous solution using 2-Hydroxypropyl-β-cyclodextrin is dependent on the specific compound of interest. Below are some references to help determine the concentration and methodology for the complexation. A lipophilic compound's ability to complex with 2-Hydroxypropyl-β-cyclodextrin is related to the compound's size, shape, and lipid partition coefficient. (Yaksh TL, Jang J, Nishiuchi Y, Braun KP, Ro S & Goodman M. Life Sci 1991;48(7):623-633) For instance, capsaicin, a lipid-soluble compound, was dissolved in 20% 2-Hydroxypropyl-β-cyclodextrin aqueous solution which resulted in a shift of the lipid partition coefficient from 0.01 to 2.05. According to the Research Journal of Pharmaceutical, Biological and Chemical Sciences: "Generally, guest /cyclodextrin complexes have a molar ratio, [guest]/[cyclodextrin] of 1:1. The molar ratio can be higher or lower and this depends on the size of the guest molecule and the identity of the cyclodextrin. Many techniques which are used to form CD-complexes are kneading, co-precipitation, dry mixing, sealing, slurry complexation, neutralization, spray drying, freeze-drying, and solvent evaporation."
