1alpha-Hydroxyvitamin D3 is a synthetic analog of Calcitiol (the hormonal form of vitamin D3), which shows identical potency with respect to stimulation of intestinal calcium absorption and bone mineral mobilization. It is a vitamin D source that has a high affinity for the vitamin D receptor (VDR) and is more potent than natural vitamin D. 1alpha-Hydroxyvitamin D3 binds to the VDR to form a complex, which then binds to vitamin D response elements (VDREs) in the promoter regions of vitamin D-responsive genes. This binding activates transcription of these genes, resulting in a variety of biological effects. These effects include the regulation of calcium and phosphate homeostasis, the stimulation of bone formation, and the modulation of immune responses. 1alpha-Hydroxyvitamin D3 also has anti-inflammatory and anti-cancer activities, and may act to have neuroprotective effects.
