11β-HSD2 Antibody (C-9) is a mouse monoclonal IgG1 kappa light chain antibody that detects 11β-HSD2 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). 11β-HSD2 (C-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. 11β-HSD2 plays a vital role in regulating glucocorticoid hormone action within target tissues by catalyzing the conversion of active corticosteroids, such as cortisol, into inactive forms, like cortisone. This enzymatic activity is particularly important in tissues where the balance of glucocorticoids and mineralocorticoids is critical for maintaining physiological homeostasis. 11β-HSD2 is predominantly expressed in the placenta and in aldosterone target tissues, including the kidney, parotid gland, colon, and skin, where 11β-HSD2 helps prevent inappropriate activation of mineralocorticoid receptors by cortisol. Dysregulation of 11β-HSD2 has been implicated in various conditions, including hypertension and metabolic disorders, highlighting 11β-HSD2′s significance in both development and disease. 11β-HSD2′s ability to modulate receptor occupancy by glucocorticoids underscores its potential as a therapeutic target in disorders related to glucocorticoid signaling.

Is this antibody tested for western blot in mouse samples?

Thank you for your question. Based on our tests with the product, 11β-HSD2 Antibody (C-9): sc-365529 is only recommended for detection of 11β-HSD2 of human origin by WB, IP, IF, IHC(P) and ELISA. We do not recommend it for use on murine cell or tissue samples.

Is it possible to detect rat 11-beta-hydroxysteroid dehydrogenase type 2 with a C-9 antibody?

Thank you for your question. We have not yet confirmed reactivity with mouse and rat species for this mouse monoclonal antibody, 11β-HSD2 (C-9): sc-365529. The antibody is raised against a recombinant protein sequence that does share 78% sequence identity with the rat 11β-HSD2 protein, so there is a good chance that the antibody will react with rat. We are also aware of a recent 2016 publication citing use of the (C-9) clone in rat heart samples, please see PMID: 27614224. For a limited time, we are offering a free 10 µg sample of this antibody. What is the recommended fixation method for immunofluorescence staining with 11β-HSD2 (C-9): sc-365529 monoclonal antibody?
