NHERF-1 Antibody (6) is a mouse monoclonal IgG1 antibody that detects NHERF-1 in human samples through applications such as western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). NHERF-1, also known as EBP50, plays a crucial role in regulating the Na+/H+ exchange protein NHE3, which is essential for transepithelial sodium absorption primarily in the intestinal and renal brush border membranes. NHERF-1′s ability to interact with NHE3 through two PDZ domains allows NHERF-1 to act as a scaffold for a multiprotein complex that includes Ezrin, facilitating cAMP-mediated phosphorylation and subsequent inhibition of NHE3 activity. This regulation is vital for maintaining sodium homeostasis and fluid balance in the body. Additionally, NHERF-1′s amino-terminal PDZ domain is important for dimerization in vivo, which further influences functional interactions. The phosphorylation of NHERF-1 at Ser 289 by G protein-coupled receptor kinase 6A highlights the intricate signaling pathways that modulate activity, while NHERF-2, another member of the NHERF family, also contributes to the regulation of sodium transport through NHE2. With a significant role in cellular signaling and ion transport, NHERF-1 is a key protein for researchers studying renal and intestinal physiology.
