    NHERF-1 Antibody (4i337) is a mouse monoclonal IgG2b antibody that detects NHERF-1 in human samples through western blotting (WB) and immunoprecipitation (IP) applications. NHERF-1, also known as EBP50, plays a crucial role in regulating the Na+/H+ exchange protein NHE3, which is essential for transepithelial sodium absorption primarily in the intestinal and renal brush border membranes. Anti-NHERF-1 antibody (4i337) interacts with NHE3 through its two PDZ domains, allowing NHERF-1 to act as a scaffold for a multiprotein complex that includes Ezrin, facilitating cAMP-mediated phosphorylation and subsequent inhibition of NHE3 activity. This regulation is vital for maintaining sodium homeostasis and fluid balance in the body. Additionally, NHERF-1′s amino-terminal PDZ domain is important for its dimerization in vivo, which further influences its regulatory functions. The phosphorylation of NHERF-1 at Ser 289 by G protein-coupled receptor kinase 6A highlights the intricate signaling pathways that modulate its activity, while NHERF-1′s interaction with NHERF-2, also known as E3KARP, underscores the complexity of sodium transport regulation across different tissues.
