Neutrophil Marker를 위한 Neutrophil Elastase항체

호중구 엘라스타제 항체(G-2)는 엘라네 또는 메둘라신으로도 알려진 인간 호중구 엘라스타제(NE)의 아미노산 211-267을 인식하도록 설계된 마우스 단일 클론 IgG1 카파 경쇄 항체입니다. 항NE 항체(G-2)는 웨스턴 블롯팅(WB), 면역 침전(IP), 면역 형광(IF), 파라핀 내장 섹션을 이용한 면역 조직 화학(IHCP), 효소 결합 면역 흡착 분석(ELISA) 등의 애플리케이션을 통해 NE를 효과적으로 검출하므로 다양한 연구 환경에서 필수적인 도구로 사용됩니다. NE는 호중구의 호염성 과립에 주로 저장되는 세린 프로테아제로, 면역 반응 중에 식세포화된 병원균을 분해하고 세포 외 기질을 리모델링하는 데 중요한 역할을 합니다. 글리코실화를 포함한 NE의 번역 후 변형은 적절한 폴딩, 안정성 및 효소 활성에 중요하며, 의도하지 않은 조직 손상을 일으키지 않고 효과적인 기능을 보장합니다. 또한 이러한 변형은 보체 시스템에서 다른 단백질과의 상호작용과 혈소판 작용제로서의 역할을 조절하여 혈소판 응집, 분비 및 칼슘 동원을 향상시킵니다. 이러한 변형에 의해 유지되는 구조적 무결성은 특발성 폐섬유증, 류마티스 관절염, 성인 호흡곤란 증후군 및 낭포성 섬유증과 같은 염증성 질환에 NE가 관여하는 데 필수적입니다. NE 단일 클론 항체(G-2)는 NE의 변형된 영역을 특이적으로 표적함으로써 안정적이고 정확한 검출을 제공하여 염증 및 조직 손상의 분자 메커니즘에 대한 고급 연구를 용이하게 합니다. 아가로스, 홀스래디쉬 퍼옥시다제(HRP), 피코에리스린(PE), 플루오레세인 이소티오시아네이트(FITC) 및 여러 Alexa Fluor® 접합체를 포함한 비접합 형태와 다양한 접합 형태로 제공되는 NE(G-2) 항체는 광범위한 실험 프로토콜을 수용합니다.

Which antigen retrival method you tried with that specific antibody? Did it work with Tris-EDTA pH 9.0?

How can Neutrophil Elastase (G-2): sc-55549 monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal antibody, Neutrophil Elastase (G-2), is currently available conjugated to phycoerythrin (sc-55549 PE), fluorescein (sc-55549 FITC), Alexa Fluor® 488 (sc-55549 AF488) or Alexa Fluor® 647 (sc-55549 AF647)

Can you recommend a positive control to use with Neutrophil Elastase (G-2): sc-55549 monoclonal antibody for Western blot?

We recommend U-937 (sc-2239) or HL-60 (sc-2209) whole cell lysates to use as positive controls as we have Western blot data showing specific detection of Neutrophil Elastase in these cell lines.

Neutrophil Elastase expressed in the activated neutrophil in the lysosomes. Azurophil granules stained with alexa 488. Very good antibody recognise human and mouse neutrophil.

Antigen retrievals tested were: 1) autoclaving in citrate buffer pH 6.0; 2) protease XIV; 3) no pretreatment.
