MDR1 와 Mdr-3를 위한 MDR항체

MDR1 항체(D-11)는 아미노산 1040-1280 에피토프를 인식하여 ABCB1로도 알려진 인간 MDR1 단백질을 표적으로 삼도록 설계된 마우스 단일 클론 IgG2b 카파 경쇄 항체입니다. MDR1 단일 클론 항체(D-11)는 웨스턴 블롯팅(WB), 면역 침전(IP), 면역 형광(IF), 파라핀 매립 절편을 이용한 면역 조직 화학(IHCP), 효소 결합 면역 흡착 분석(ELISA) 등의 기술을 활용하여 마우스, 쥐, 인간 등 여러 종에서 MDR1을 효과적으로 검출하며 마우스 동형체인 Mdr-3과 교차 반응할 수도 있습니다. ABCB1, P-GP, PGY1, ABC20, CD243, ENPAT, GP170, p-170이라고도 불리는 MDR1 단백질은 ATP 결합 카세트(ABC) 수송체 계열의 중요한 구성원으로서 주로 혈액뇌장벽, 간, 신장 및 장 내 세포의 정단막에 존재합니다. MDR1의 주요 기능은 약물 수송 펌프 역할을 하여 다양한 화학 요법제와 기타 이종 생물을 세포 밖으로 활발하게 배출하는 것으로, 암 치료에서 다제내성(MDR)이 발생하는 데 중요한 역할을 합니다. One of our customers, who is working with cat # sc-55510, lot # C2415 is facing some issues with the antibody. The band of MDR1 is suppose to come at 170KDa but it is coming around 55KDa. They used1:1000 dilution in 2% BSA in Tbst. Please suggest.

Is this antibody resistant to formalin fixation and periodate oxidation? Any published reference detailing its validation (eg epitope recognized, selectivity against other transporters)? Thks in advance.

Thank you for your question. This antibody, Mdr-1 (D-11): sc-55510, is not resistant to formalin fixation as it has been successfully tested on formalin-fixed paraffin-embedded tissue sections. This data is available on our website. The antibody has been cited in over 48 publications, however we are not aware of any of these publications describing the exact epitope recognized by this clone, or experimental validation being published for cross reactivity with other transporters. Hi We are using the Mdr-1 antibody and we repeatedly see three band (in two different cell lines): 1- 130-200 kDa smear which is supposed to be the Mdr-1 band. 2 and 3- two separated narrow bands between 35-50 kDa. How could you explain that?

Thank you for your question. We do not have any information about the type of lower molecular weight bands you have described when using this Mdr-1 antibody. We would recommend first performing a secondary antibody control, where you incubate your membrane without primary antibody to determine if any of those bands may be a result of non-specific binding of the secondary antibody alone.

What is the recommended secondary reagent to be used with Mdr-1 Antibody (D-11) in Western blot?

Thank you for your question. Mdr-1 Antibody (D-11) is a mouse monoclonal IgG2b with a kappa light chain. Therefore, we would recommend using our mouse IgG kappa binding protein (m-IgGκ BP) conjugated to Horseradish Peroxidase (HRP): sc-516102. We want to order your anti MDR1 mouse antibody. The band which you show at the site looks like a smear between 130-200kDa. How could this be explained? Thank you in advance. Dina

Thank you for your question. Mdr-1 Antibody (D-11): sc-55510 is a mouse monoclonal antibody which recognizes Mdr-1 of human origin by Western blot. The Mdr-1 protein has a predicted molecular weight of 170 kDa, however it has been described as hyperglycosylated and can appear as a diffuse band by Western blot (reference PMID: 12138126). This antibody is very popular and has been cited in over 40 publications, primarily for use in Western blot. A list of these product citations is available on our website for this antibody.
