CAP-18 와 LL-37를 위한 LL-37 & CAP-18항체

LL-37 항체(D-5)는 인간 유래의 LL-37 단백질을 WB, IP, IF, IHC(P) 및 ELISA로 검출하는 IgG1 κ 마우스 단일 클론 LL-37 항체(카탈리시딘 전구체 유도체 LL-37 항체, 대식세포/다형핵 백혈구(PMN) 특이 리소좀 국소화 펩티드 LL37 항체 또는 CAP-18(CAMP) 전구체 후 번역 절단산물 LL37 특이 항체로도 지정됨)입니다. LL-37 항체(D-5)는 비접합 항-LL-37 항체 형태와 아가로스, HRP, PE, FITC 및 여러 Alexa Fluor® 접합체를 포함한 여러 접합 형태의 항-LL-37 항체 형태로 제공됩니다. 카텔리시딘은 호중구의 퍼옥시다제 음성 과립에서 발견되는 항균 단백질의 일종입니다. 카테리시딘은 디펜신으로 알려진 단백질 계열과 함께 미생물의 국소 감염과 전신 침입을 방지하여 1차 방어선에 참여합니다. CAP-18 전구체(FALL-39 펩타이드 항생제, 양이온성 항미생물 단백질, CAMP, HSD26)는 항균 펩타이드 LL-37(아미노산 134-170)을 포함하는 카탈리시딘 항미생물 단백질입니다. 시스테인이 풍부한 펩타이드인 디펜신은 β 주름 형태로 접히는 반면, LL-37은 시스테인이 없는 펩타이드이며 양면성 α 나선형 형태를 취할 수 있습니다. LL-37은 박테리아 지질다당류(LPS)에 결합하며 염증 부위에 비만 세포를 모집하는 강력한 화학 주성 인자입니다. LL-37은 건선, 아급성 홍반성 루푸스, 피부염 및 니켈 접촉 과민증을 포함한 염증성 피부 질환에 존재합니다. 정상적인 피부 표피에서는 발견되지 않습니다. 분비된 단백질은 주로 골수, 고환 및 호중구에서 발현됩니다. 생쥐와 쥐의 상동 단백질인 CRAMP(카테린 관련 항균 펩타이드)도 카테리시딘 계열의 숙주 방어 펩타이드에 속합니다. 여기에는 다양한 미생물 병원체에 대한 항균 활성을 유도하고 상처 회복 중에 중간엽 세포를 활성화하는 강력한 항균 펩타이드의 전구체가 포함됩니다. We are using this Ab (LL-37 (D-5): sc-166770) along with m-IgGk-BP-HRP (2ndry Ab as recommended) to analyze protein expression in cancer cell lines (positive control HeLa cells), but having difficulty to see any signal. Any suggestions? thanks

Thank you for your question. We recommend using this primary antibody at a starting dilution of 1:200 and loading 40-60 µg of protein. A link to our complete recommended protocol for Western blot is provided below. LL-37 is a short secreted peptide (~4.5 kDa) that is difficult to detect in Western blot. We have not tested this antibody in HeLa cells, so we do not know how abundant LL-37 is in this cell line. We are aware of publications citing use of this antibody in Western blot and we would recommend reviewing these references for more detailed information about using the antibody (e.g., PMID: 25330301 & 23940207). Can this mouse primary antibody be used for IF or IHC staining in mouse tissue?

Yes. LL-37 Antibody (D-5): sc-166770, is available conjugated to HRP (sc-166770 HRP), 200 µg/ml, for WB, IHC(P) and ELISA; phycoerythrin (sc-166770 PE), fluorescein (sc-166770 FITC), Alexa Fluor® 488 (sc-166770 AF488) or Alexa Fluor® 647 (sc-166770 AF647), 200 µg/ml, for IF, IHC(P) and FCM.
