글리코포린 A 항체(R10)는 웨스턴 블롯팅(WB), 면역 침전(IP), 면역 형광(IF), 파라핀 내장 섹션을 이용한 면역 조직화학(IHCP) 및 유세포 분석(FCM)으로 인간 유래의 글리코포린 A를 검출하는 마우스 단일 클론 IgG1 카파 경쇄 항체입니다. 항 글리코포린 A 항체(R10)는 비접합 형태와 아가로스, 홀세라디시 퍼옥시다제(HRP), 피코에리스린(PE), 플루오레세인 이소티오시안산염(FITC), 다중 Alexa Fluor® 접합체를 포함한 다양한 접합 형태 모두에서 사용할 수 있습니다. 글리코포린 A는 인간 적혈구 표면에 위치한 중요한 시알로당 단백질로 적혈구의 구조적 완전성을 유지하고 면역 체계 상호 작용을 촉진하는 데 중요한 역할을 합니다. 글리코포린 A는 세포막을 한 번 가로질러 세포 외 환경에 아미노 말단을 제시하며 혈액형 항원성 기능에 필수적인 역할을 합니다. 글리코포린 A, B, C를 포함한 글리코포린 표면 항원의 유전적 다양성은 혈액형 표현형을 결정하므로 이러한 단백질은 수혈 호환성 및 용혈성 질환을 이해하는 데 필수적입니다. 4q31.21 염색체에 위치한 글리코포린 A 유전자는 7개의 엑손으로 구성되어 있으며 91개의 아미노산 단백질을 코딩하는 글리코포린 B와 높은 상동성을 보입니다. 글리코포린 A의 구조와 기능을 이해하는 것은 혈액형 항원 발현과 혈액 관련 질환의 잠재적 치료 표적에 대한 통찰력을 제공하여 임상 응용 및 적혈구 생물학 연구에 매우 중요합니다.

Could you please share the protocol of the IHC staining of the second image that you posted? My humanized mice samples consist of both human and mouse rbc, will this cause any unspecific binding to mouse rbc?

Thank you for your question. A link to our recommended protocol that was used for IHC staining with this Glycophorin A Antibody (R10): sc-53905 is provided below. This antibody is recommended for detection of Glycophorin A of human origin. Can Glycophorin A (R10): sc-53905 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, Glycophorin A (R10): sc-53905 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 3 out of 5 by from Positive Results in WBWe tested this antibody in WB against red blood cell lysate and we saw a strong and clear signal at 37 KDa. Otherwise, in more diluted samples the antibody didn't work.

Rated 2 out of 5 by from WBWe saw band for Glycopurin A (R10) in the WB of mouse lung primary fibroblast, but it was in the wrong fraction. Instead of membrane fraction, the band was in cytoplasmic fraction. We had negative and positive control for the fraction extraction. The size of the band also was not true, it was about 50 KD. We used anti-mouse IgG1.
