FIH-1 Antibody (A-5) is a mouse monoclonal IgG2a kappa light chain antibody that detects FIH-1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). FIH-1 (A-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Factor Inhibiting HIF-1 (FIH-1) plays a crucial role in cellular response to hypoxia by regulating hypoxia-inducible factor 1 alpha (HIF-1α) activity. FIH-1 functions as an asparaginyl hydroxylase, catalyzing the hydroxylation of asparagine residue 803 within the carboxy-terminal transactivation domain of HIF-1α, which prevents interaction with co-activators and thus inhibits transcriptional activity. This regulatory mechanism is vital for maintaining cellular homeostasis under varying oxygen levels, as dysregulation of HIF-1α can lead to pathological conditions such as cancer and ischemia. FIH-1 also interacts with von Hippel-Lindau (VHL) tumor suppressor protein, further contributing to HIF-1α activity repression. In transiently transfected human osteosarcoma cells, FIH-1 is localized in the cytoplasm, and FIH-1 structure features a jellyroll-like β-barrel containing ferrous-binding triad residues, essential for enzymatic function. Human FIH-1 gene is located on chromosome 10q24, highlighting significance in genetic regulation and potential implications in disease.
