Its activity largely pivots on the phosphorylation state of its partnering subunit, eIF2α. The ensemble of chemicals dubbed eIF2S3 Activators indirectly modulate the protein's activity through their interactions with various cellular pathways that influence eIF2α's phosphorylation. Notably, chemicals like Salubrinal and Sephin1, by inhibiting eIF2α dephosphorylation, can pave the way for increased eIF2S3 function.

In contrast, chemicals such as ISRIB, by enhancing eIF2B activity, counteract the inhibitory effects of eIF2α phosphorylation, thus indirectly facilitating eIF2S3's role in protein synthesis initiation. Moreover, ER stress inducers like Tunicamycin and Thapsigargin trigger pathways that influence eIF2 signaling, and by extension, eIF2S3's function.
