Nestin Antibody (2Q178) is a mouse monoclonal IgG1 antibody that detects nestin in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). Nestin is a crucial intermediate filament protein predominantly expressed in embryonic central nervous system progenitor cells, where nestin plays a vital role in maintaining the structural integrity and function of these cells during development. Nestin′s dynamic presence in the intermediate filament network is essential for muscle development, as nestin polymerizes with other proteins such as Desmin and Vimentin, forming heterodimer coiled-coil structures that contribute to the formation of 10 nm intermediate filaments. Nestin′s localization is particularly important; for instance, nestin deletion in the intermediate filament consensus rod domain can disrupt localization in central nervous system precursor cells and radial glial cells, which may have significant implications for neurodevelopmental processes. Furthermore, nestin serves as a marker for neuroepithelial stem cells, glioma cells, and tumor endothelial cells during periods of rapid growth, highlighting nestin′s relevance in cancer biology. During the differentiation of neurons, nestin is localized to growth cones, suggesting a potential role in guiding axon elongation. Additionally, in the rat adrenal gland, nestin expression is observed in the zona fasciculata and zona reticularis, as well as in dermatomal cells and myoblasts during the early stages of myogenesis, underscoring nestin′s diverse functional roles across different tissues.
