Na+/K+-ATPase α Antibody (M7-PB-E9)は、Na+/K+-ATPase α サブユニットを認識するように特異的に作製されたマウスモノクローナル IgG1 κ軽鎖抗体です。Na+/K+-ATPase α モノクローナル抗体（M7-PB-E9）は、ヒツジ由来の Na+/K+-ATPase α タンパク質に対して作製されており、ウェスタンブロッティング（WB）、免疫沈降（IP）、免疫蛍光（IF）、免疫組織化学などの応用により、マウスおよびヒト由来のαアイソフォーム、ならびにラット由来のα3アイソフォームを効果的に検出します。さらに、Na+/K+/K+-ATPase α (M7-PB-E9) 抗体は、ウシ、ブタ、イヌを含む他の動物種と反応性を示しますが、Xenopus やニワトリの α1 アイソフォーム、ラットの α2 または α3 アイソフォームとは交差反応しません。Na+/K+-ATPaseはα、β、γサブユニットからなる重要な膜結合酵素であり、3つのナトリウムイオンを細胞外に、2つのカリウムイオンを細胞内に積極的に輸送することにより、必要不可欠な電気化学的勾配を維持する役割を担っている。この活性は、細胞容積の調節、静止膜電位の維持、他の溶質の二次的な能動輸送の促進など、様々な細胞プロセスに不可欠である。αサブユニットは触媒コアとして機能し、ATPとイオンの結合部位を持ち、βサブユニットは適切なフォールディングと膜局在化を保証し、γサブユニットは酵素のATPに対する親和性を調節する。Na+/K+-ATPaseは、細胞骨格と結合するアンキリンやシグナル伝達経路に関与するSrcキナーゼなど、いくつかの制御タンパク質と相互作用する。これらの相互作用は酵素の制御と細胞内シグナル伝達機構に不可欠である。Na+/K+-ATPase α (M7-PB-E9)モノクローナル抗体は、イオン輸送ダイナミクス、細胞恒常性、生理学的プロセスを支配する複雑なタンパク質相互作用を研究する研究者にとって不可欠なツールであり、複数の生物種や実験手法にわたって信頼性の高い検出を提供します。

Is it possible to use the Ab for IF staining of fixed cells? If yes, what is the suitable Ab concentration to use? Thank you, Gaia

Thank you for your question. Na+/K+-ATPase α Antibody (M7-PB-E9): sc-58628 is recommended for detection of all α isoforms of Na+/K+-ATPase of mouse and human origin and Na+/K+-ATPase α3 of rat origin by WB, IP, IF and IHC(P), so yes, it is suitable for what you describe. The appropriate antibody dilution can be found in the product datasheet: starting dilution 1:50, dilution range 1:50-1:500.

Does this antibody suitable for fish species?

Can you recommend a positive control to use with Na+/K+-ATPase α (M7-PB-E9): sc-58628 monoclonal antibody for Western blot?

We recommend HeLa (sc-2200) or Hep G2 (sc-2227) whole cell lysates or human brain (sc-364375) or human kidney (sc-363764) tissue extracts to use as positive controls as we have great Western blot data showing specific detection of Na+/K+-ATPase α in these cell lines.

How can Na+/K+-ATPase α (M7-PB-E9): sc-58628 mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. Na+/K+-ATPase α (M7-PB-E9): sc-58628 monoclonal antibody is currently available conjugated to phycoerythrin (sc-58628 PE), fluorescein (sc-58628 FITC), Alexa Fluor® 488 (sc-58628 AF488) or Alexa Fluor® 647 (sc-58628 AF647)

Note that our own test was in pig cells, and depending on the protein, it is common for antibodies to show some species specificity. We have not tested in other species.
