CD71 Antibody (YTA 74.4) is a rat monoclonal IgG2a antibody that recognizes CD71 protein of mouse and rat origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). CD71 (YTA 74.4) antibody is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CD71, also known as transferrin receptor (TFR), is a type II membrane glycoprotein essential for iron homeostasis through receptor-mediated endocytosis. This process supports DNA synthesis and cellular proliferation, as iron serves as a crucial cofactor for many enzymes. CD71 shows high expression in proliferating cells, reticulocytes, and erythroid precursors, with increased levels in activated lymphocytes, monocytes, and macrophages during immune responses. CD71 expression extends to most dividing cells and brain endothelium, indicating broad physiological significance. A second transferrin receptor, TFR2, also facilitates transferrin-bound iron uptake, primarily in the liver, contributing to iron regulation. TFR2 gene mutations can cause hereditary hemochromatosis type III (HFE3), an iron overload disorder common among Caucasians, highlighting CD71′s importance in iron metabolism and related diseases.

Can CD71 (YTA 74.4): sc-59112 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, CD71 (YTA 74.4): sc-59112 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 4 out of 5 by from CD71 in rat liver WCLConditions: 60 ug protein/lane, 7.5% gel, BME dye, no heat. Primary antibody, 1:1000 in 2.5% nonfat milk-TBST; incubated overnight at 4C. Secondary antibody, 1:10k in 2.5% nonfat milk-TBST; incubated for 1 hr at RT. Imaged using ChemiDoc imager system and femto substrate. Strong band just below 100 kDa. There is some smearing, but heating samples at 37C for 30min showed reduced smearing; further optimization may lead to a cleaner blot.
