ADD1 encodes adducin α, a membrane skeletal protein that caps actin filaments and promotes spectrin–actin network assembly at the cell cortex. Through regulation of cytoskeletal remodeling, adducin α influences cell shape, adhesion, and membrane stability, with downstream effects on processes such as cell migration and mechanotransduction. Adducin function is modulated by phosphorylation-dependent signaling, including PKC- and PKA-linked pathways that alter actin-binding dynamics and cortical organization. Dysregulation of cytoskeletal and membrane scaffolding proteins, including ADD1, has been associated with altered epithelial barrier properties and cellular phenotypes relevant to cardiovascular and neurobiological research.
Adducin α CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the ADD1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the ADD1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the ADD1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Adducin α protein expression.
This CRISPR knockout system enables efficient generation of ADD1-deficient cell models for investigation of Adducin α signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
