SIMPL, which stands for Signal Integration by the Mitogen-activated Protein kinase (MAPK) Pathway, is a protein that in humans is encoded by the SIMPL gene. This protein acts as a modulator of transcriptional response within the MAPK signaling pathway, which is a critical cascade in cellular biology that transmits molecular signals from the cell membrane to the nucleus.The MAPK pathway is involved in a wide array of cellular processes, including growth, differentiation, and response to stress. It is made up of a series of kinase enzymes that activate one another in a specific sequence, leading to the phosphorylation and activation of downstream targets, including transcription factors that regulate gene expression.

SIMPL itself is not a kinase but rather a nuclear protein that interacts with the MAPK pathway components. It has been found to function as an adaptor or scaffold protein, facilitating the interaction between the MAPKs and their target transcription factors in the nucleus. Specifically, SIMPL has been shown to interact with tumor necrosis factor-alpha (TNF-alpha) and modulate the activity of transcription factor NF-kappaB, which plays a pivotal role in immune and inflammatory responses.By influencing the NF-kappaB pathway, SIMPL can impact the transcription of genes that are involved in immune and inflammatory responses, cell survival, and apoptosis. Dysregulation of NF-kappaB has been linked to various pathological conditions, including cancer, inflammatory and autoimmune diseases, and viral infections, which highlights the potential significance of SIMPL in disease processes.
