Reep2 encodes receptor expression-enhancing protein 2 (REEP2), a member of the REEP family implicated in shaping endoplasmic reticulum membranes and supporting ER–microtubule interactions that influence intracellular trafficking. REEP2 contributes to membrane protein handling and neurite-associated ER organization, processes important for maintaining neuronal morphology and proteostasis. Disruption of REEP family functions has been linked to axonal vulnerability and neurodegenerative mechanisms, making Reep2 a relevant target for studying pathways that impact axon maintenance and stress responses. In mouse systems, Reep2 perturbation can be used to interrogate ER remodeling, organelle dynamics, and trafficking-dependent signaling in neurons and other secretory cell types.
REEP2 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Reep2 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Reep2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Reep2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish REEP2 protein expression.
This CRISPR knockout system enables efficient generation of Reep2-deficient cell models for investigation of REEP2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
