PP2Cκ Antibody (E-8) is a mouse monoclonal IgG1 antibody that detects PP2Cκ in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). PP2Cκ, also known as Protein phosphatase 2C isoform kappa, is a crucial 372 amino acid protein located in the mitochondrial matrix, where PP2Cκ plays a vital role in regulating mitochondrial membrane permeability transition pores. This regulation is essential for maintaining cell survival, cardiac function, and proper embryonic development. PP2Cκ′s importance is underscored by knockdown studies leading to cell death due to the loss of mitochondrial membrane potential, highlighting PP2Cκ′s protective role in cellular health. Additionally, PP2Cκ interacts specifically with the branched-chain-alpha-ketoacid dehydrogenase (BCKD) complex, facilitating the dephosphorylation of Ser293, which ultimately inhibits branched-chain amino acid metabolism. PP2Cκ shows highest expression levels in the brain, diaphragm, and heart, and exists in three isoforms produced by alternative splicing, further emphasizing PP2Cκ′s functional diversity and significance in various biological processes.
