PLXNB1 encodes plexin-B1, a single-pass transmembrane receptor for class 4 semaphorins that coordinates cytoskeletal remodeling and directional cell movement. Through coupling to small GTPase signaling modules, including Rho family pathways and Ras-related inputs, plexin-B1 regulates actin dynamics, adhesion turnover, and neurite guidance. Plexin-B1 activity intersects with receptor tyrosine kinase and integrin-associated signaling to influence migration and invasive behavior across multiple cell contexts. Dysregulated PLXNB1 expression or signaling has been implicated in neurodevelopmental processes and in cancer-associated phenotypes such as altered motility and metastatic potential, supporting its use as a mechanistic node in pathway studies.
plexin-B1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the PLXNB1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the PLXNB1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the PLXNB1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish plexin-B1 protein expression.
