Npdc1 encodes NPDC-1, a neuronal and glial differentiation-associated protein implicated in the control of cell cycle progression and maturation programs within the nervous system. NPDC-1 has been linked to regulation of proliferation-to-differentiation transitions, with reported interactions that influence transcriptional outputs and growth control pathways in neural lineages. In mouse models and neural cell systems, altered NPDC-1 activity is used to interrogate mechanisms of neurite outgrowth, neuronal plasticity, and lineage specification. Dysregulation of these processes is relevant to neurodevelopmental abnormalities and to cellular states associated with aberrant proliferation in nervous tissue.
NPDC-1 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Npdc1 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Npdc1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Npdc1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish NPDC-1 protein expression.
This CRISPR knockout system enables efficient generation of Npdc1-deficient cell models for investigation of NPDC-1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
