API-1 is a PKB inhibitor. The compound acts by halting Akt membrane translocation through binding the pleckstrin homology domain of Akt. API-1 also inhibits EGF-induced kinase activity of Akt1, AKT2 and AKT3. Activity results in cell growth arrest and apoptosis in human cancer cells with constitutively active Akt. Both in vivo and in vitro antitumor activity has been observed along with cell arrest and apoptosis in cancer cells with constitutive Akt activity.
