TAPI-0 is a hydroxamate-based inhibitor of collagenase, gelatinase, and TACE (TNF-a converting enzyme/ADAM17 IC50 = 100 nM). TAPI-0 blocks spontaneous and PMA-induced release of TNF-a in transfected COS 7 cells. TAPI-0 blocks the maturation of cytokines, soluble cytokine receptors, and other proteins that require the action of proteases.
