SEE ALSO...

NALP7 Antibody (C-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects NALP7 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). NALP7 (C-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NALP7, also known as PYPAF3, plays a critical role in regulating inflammatory responses by inhibiting interleukin-1 beta (IL-1β) secretion through direct interaction with caspase-1. This function is particularly important as IL-1β is a key mediator in the inflammatory process, and dysregulation can lead to various inflammatory diseases. NALP7 is predominantly expressed in tissues such as the uterus and ovary, with lower expression levels in the heart and brain, indicating potential involvement in reproductive health and inflammatory responses in these tissues. Mutations or defects in NALP7 are associated with hydatidiform moles, which are abnormal pregnancies characterized by cystic degeneration of placental villi and absence of an embryo. Studies have shown that silencing NALP7 via RNA interference can inhibit carcinoma cell line growth, suggesting a significant role in cell proliferation and cancer biology. NALP7 is located on chromosome 19q13.42, within a cluster of other NALP genes, highlighting evolutionary significance and potential functional redundancy within this gene family.

I saw you showed the band in F9 cell lysate, but NLRP7 does not exit in mouse genome. Does this antibody cross interact with NLRP2?

Asked by: Huihan
Thank you for your question. The data with mouse lysates has been flagged for removal, but unfortunately has not yet been pulled from the website. We do not support this antibody for detection of any NALP family member in mouse. The homology for NALP2 of mouse origin is very low for the immunogen used (34% over 106 amino acids) however it does contain two regions that have enough homology that there is the possibility of cross-reactivity. Without further characterization however we will not guarantee this reactivity. Please contact Technical Service by phone, (800)-457-3801 option 2, email <scbt@scbt.com>, or by live chat directly on our website, www.scbt.com if you have any further questions.

Hi, I want to order a NLRP7 antibody (N-20: sc-50642), but it seems no longer available. Is the C-8 (sc-377190) targeting the same region on NLRP7 as the N-20? Does it work well?

Asked by: Huihan
Thank you for your question. Our NLRP7 sc-377190 is raised against amino acids 16-170 of NALP7 of human origin, which encompasses the immunogen used for sc-50642. This antibody performs very well in house for Western Blot, IF and IHC including paraffin embedded sections.

Can NALP7 (C-8): sc-377190 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: Dr Ninau Qelp
Thank you for your question. Yes, NALP7 (C-8): sc-377190 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
